PROJECT SEQUENCING SERVICE

This exclusive service includes all steps necessary to determine the complete double- or single-stranded sequences of large DNA fragments (up to 30 kb). A convenient option.

BaseClear's project sequencing service includes:

• If necessary purification of DNA samples
• DNA concentration measurement
• Design and synthesis of all necessary primers 
• Optimal sequencing reaction, if necessary under specific conditions (e.g. in case of GC-rich DNA) 
• Assembly and (manual) editing of the sequence data 
• Comparison to a reference sequence if available
• Customer will be informed regularly about the progress 
• Extensive advice for special projects 
• Delivery time depends on read length and sequencing strategy. In case of primer walking we analyze > 1 kb per week.

Single- or double-stranded sequencing
Sequencing can be performed for one or for both DNA strands. Double-stranded DNA sequencing results in a higher accuracy of the sequence data. If a reference sequence is known, BaseClear can design the necessary primers on the basis of this.

Primer walking
If a reference sequence is not available, or if a clone contains unknown inserts, the DNA can be sequenced via primer walking. Due to the overlap of the resulting sequences, the complete contiguous DNA sequence can be determined.
