special sequencing service


As a specialist in DNA sequencing, BaseClear has developed special efficient protocols for sequencing samples that cannot be sequenced easily by straightforward normal sequencing.

Large construct sequencing

Sequencing of large DNA constructs such as BAC, PAC, cosmid or lambda DNA is complicated due, for example, to the large parts of non-coding DNA. BaseClear has developed a special protocol for these types of DNA. Our service includes a DNA quality check, data editing and a rerun if necessary. The sequence reactions have a read length up to 400 bases. 

Genomic sequencing

For genomic DNA, we offer a special sequencing service. Our service includes a single sequencing reaction on genomic DNA (< 5 Mb), including DNA quality check, data editing and a rerun if necessary. The read length is variable.

siRNA sequencing

BaseClear can sequence siRNA constructs via runs from both sides of the siRNA, using two different sequencing kits. siRNA inserts will be sequenced under standard conditions and under conditions for GC-rich and hairpin regions. Both a forward and reverse short run (max 500 nucleotides) will be performed. Our service includes a DNA quality check, data editing and a rerun if necessary. Upon request, the resulting sequences will be assembled into a contig sequence.

Note: some parts of the genome are almost impossible to read through, due to high stability regions, secondary structures, mispriming of primers, etc. Also certain siRNA inserts are almost impossible to read through, due to the high stability of the hairpin structure.
