Methylation Kits
DNA methylation involves the addition of a methyl group to cytosines in DNA that constitute CpG dinulcleotides catalysed via DNA methyltransferase enzymes within the cell.
Cluster of CpG dinucleotides are called CpG “island” and are irregularly distributed throughout the genome. Abnormal methylation of CpG islands is often related to diseases such as cancer. Studying DNA methylation patterns in some cancers has led to new methods used in cancer diagnosis, prognosis and treatment. DNA methylation in also being recognized as an important factor influencing gene expression in the aging process as well as in heritable diseases.
Methylation detection
BaseClear offers the EZ DNA Methylation and EZ DNA Methylation Gold kits, for precise, rapid DNA methylation detection. Both kits are based on a three step reaction process between cytosine and sodium bisulfite resulting in cytosine being converted into uracil. The EZ DNA Methylation Gold kit is a refinement of the EZ DNA Methylation kit in that it integrates the DNA denaturation and bisulfite conversion process into one step. This means the entire procedure only takes 5-6 hours. Both products have been designed to minimize template degradation, loss of DNA during treatment and clean-up, and to provide complete conversion of unmethylated cytosines. Recovered DNA is ideal for PCR amplification for downstream analyses including restriction endonuclease digestion, sequencing, microarray’s, etc.
Methylated standards
The Universal Methylated DNA and Universal Methylated Human DNA Standards include enzymatically methylated DNAs together with specially designed primer sets to be used in conjunction with EZ DNA Methylation and EZ DNA Methylation Gold kits to assess the efficiency of bisulfite mediated conversion of DNA.
