Isolation and handling of ribonucleic acid (RNA) has historically been thought to be more difficult than DNA due to the presence of ubiquitous and stable RNases. However, this issue can be remedied if the correct tools are used. Purification of high-quality, intact RNA from biological samples or in vitro reactions is important to the success of all downstream applications. To achieve reproducible, trustworthy results from quantitative RT-PCR, Northern blots, microarrays, and cDNA library prep for Next-Generation sequencing, it is important to use the best RNA purification methods available.
With the rigors of RNA research in mind, the scientists at Zymo Research have developed special products that effectively stabilize and preserve RNA in samples prior to purification, as well as a comprehensive portfolio of kits for the purification of high-quality, inhibitor-free RNA from essentially any sample source. All purification methods feature innovative Zymo-Spin™ columns to ensure low-volume elution of ultra-pure RNA that is free of salts and inhibitors. For most sample sources, Zymo Research offers both organic- (TRI Reagent/TRIzol) and non-organic-based methods, allowing the researcher to select the purification method they prefer. While most kits are designed for the isolation of total RNA, they are also efficient at recovering small RNAs (≥17 nucleotides). Several kits also feature unique protocols for the selective enrichment of either small RNA (<200 nucleotides), large RNA (>200 nucleotides), or both small and large RNAs separately in different fractions, at the discretion of the user.
RNA may need to be cleaned up to remove enzymes, buffers, or chemical inhibitors and concentrated for use in certain applications. Depending on the sample type and downstream application, Zymo Research provides a variety of ribonucleic acid (RNA) purification kits for the isolation of high-quality, DNA-free RNA. Whether it’s total RNA including small RNAs, separate fractions of RNA (large RNA >200 nt or small RNAs 17-200 nt), or RNA and DNA from the same sample, there is a technology specifically designed for the application. Researchers can isolate RNA from a wide variety of sample types including fresh/frozen cells and tissues, FFPE tissues, whole blood, plasma, serum, urine, yeast, bacteria, plant tissue, and more. For many downstream applications, it is essential to have RNA that is free of contaminating genomic DNA. Zymo Research's RNA isolation kits remove the vast majority of genomic DNA and feature convenient in-column DNase I treatment. The DNA-Free RNA Kit™, Direct-zol™ RNA and Quick-RNA™ kits include DNase I for fast and efficient genomic DNA removal.
Small & microRNA Isolation
The importance of miRNAs and other small RNAs in regulating gene expression and other cellular processes is clear, but many kits from other companies do not effectively recover miRNAs and other small RNA molecules. In addition, many small RNAs are expressed at low abundance, or only in specific cell types. To obtain results you can trust, you need to recover ALL small RNAs during your purification. The RNA isolation kits from Zymo Research efficiently recover small RNA molecules as short as 17 nucleotides.
RNA and DNA can be isolated simultaneously or into two separate fractions. Purify up to 25 µg DNA and/or 50 µg RNA with DNA/RNA co-purification and parallel purification kits from Zymo Research.
The ZR small-RNA™ Ladder consists of four single-stranded RNA oligonucleotides 17, 21, 25, and 29 bases in length and is suitable as a molecular weight marker for microRNAs in polyacrylamide gel electrophoresis (PAGE) experiments. The RNA ladder is supplied in water and can be stained with dyes specific for single-stranded nucleic acid species (e.g, GelStar®).
In many cases high-quality enzymes are needed to increase the efficiency of RNA purification methods. DNase I is suited for applications including removal of DNA template after in vitro transcription, and removal of DNA from RNA samples prior to applications such as RT-PCR. Proteinase K is a stable serine protease with broad substrate specificity that will degrade many proteins in their native conformation even in the presence of detergents (e.g., SDS). RNase A specifically cleaves at the 3′-side of pyrimidine (uracil or cytosine) phosphate bonds and is compatible for use in RNase protection assays and the analysis of the structure of RNA sequences in RNase structure mapping experiments.
High-throughput RNA purification and automated RNA extraction methods are now required in many different types of experiments. Zymo Research offers kits available in two 96-well formats (i.e., Zymo-Spin™ plate and magnetic bead). The magnetic bead format is completely adaptable for use with automated liquid handlers and robotics. Sample Collection & Stabilization
PRNA is susceptible to degradation by exposure to ubiquitous ribonucleases, elevated temperatures, pH extremes, and other harsh conditions. The stabilization of intact RNA is a key factor that determines the quality and consistency of downstream RNA isolation and analyses. Improper sample collection and storage can dramatically and irreversibly compromise the integrity of RNA.
